Rapamycin (Sirolimus)
12 capsules/bottleRapamycin is considered to be one of the most promising healthspan-promoting therapies for its role in slowing down the formation of senescent cells through its inhibition of the mTOR complex I pathway.
Taken in small doses over an extended period of time, Rapamycin supports processes that preserve cellular health with age, leading to a longer period of life spent active and with less dysfunction.

Ongoing Care
We will schedule a lab draw & follow-up visit after the 3rd month of treatment. After that, quarterly check-ins with annual lab draws.
"I believe that the benefits of low dose Rapamycin likely outweigh the risks for many people and that it might be worth starting earlier vs later, given that in mouse studies the best benefits happened when started at 9 months (equivalent to 30 years in humans)."
Amy Killen, MD
Medical AdvisorWe prescribe sirolimus because it is more bioavailable than the compounded version of rapamycin.
Generally, 5mg capsules taken once a week orally. New patients will be titrated up by clinician guidance. Providers may also cycle patients off for a short period of time.
We require a lab to start and then after 3 months of treatment. After that we test annually.
Possible side effects include sore or inflamed mouth, weakened immune system, anemia or fatigue, constipation or diarrhea, elevated lipid levels, high blood pressure, swollen hands or feet. Please contact us on the onset of any symptom you think may be related to treatment.
Rapamycin directly stimulates the cellular cleaning process known as autophagy, effectively clearing toxic debris and promoting cellular rejuvenation.
Cellular senescence is a process that results from a variety of stresses and leads to a state of irreversible growth arrest. Senescent cells accumulate during aging and have been implicated in promoting a variety of age-related diseases.
The mTOR pathway, found in all cells, determines when a cell should grow and divide. However, with time, it can become too active, resulting in unchecked cell growth and compromised tissue health. Rapamycin is a direct inhibitor of the mTOR pathway. Low doses of rapamycin appear to reset mTOR activity to a more youthful state—active when needed and dormant when not required.
